TOS Propionate Agar Base (Tos-Mup Agar) is a selective medium used for the enumeration of presumptive Bifidobacteria in milk products and yogurt. Formulated in accordance with ISO 29981 (IDF 220), it is designed to allow for the specific count of bifidobacteria while inhibiting the growth of symbiotic lactic acid bacteria such as Lactobacillus and Streptococcus species.

This medium utilizes TOS (Galacto-oligosaccharide) as the primary carbohydrate source, which is highly preferred by Bifidobacterium species. The Peptone from Casein and Yeast Extract provide the essential nitrogen, vitamins, and minerals required for growth. Sodium Propionate acts as a selective agent to suppress the growth of competitive flora. Selectivity is further enhanced by the aseptic addition of the antibiotic Mupirocin. Because Bifidobacteria are obligate anaerobes, the medium is designed for use with anaerobic incubation techniques. The combination of TOS and Mupirocin (TOS-MUP) provides a reliable method for distinguishing bifidobacteria from the high background of other probiotic bacteria often found in dairy formulations.
