The differentiation is dual-fold: Bromothymol Blue acts as a pH indicator (turning yellow when lactose is fermented), and the addition of TTC (Triphenyltetrazolium Chloride) allows for the identification of organisms that reduce TTC to red formazan. E. coli and coliforms typically appear as yellow colonies with a yellow halo, while non-lactose fermenters remain blue or green.
