TTC Chapman Agar, also known as Lactose Agar TTC with Tergitol 7, is a selective and differential medium used for the detection and enumeration of Escherichia coli and coliform bacteria in water samples. It is formulated in accordance with the ISO 9308-1:2000 standard (previous version) and is particularly effective for the examination of water using the membrane filtration technique.

This medium is designed to provide rapid presumptive results for coliforms by combining selective inhibition with clear color differentiation. The Peptone Mixture and Beef Extract provide the essential nitrogen, vitamins, and minerals. Lactose is the fermentable carbohydrate. The selectivity is primarily due to Tergitol 7 (Sodium Heptadecyl Sulfate), which strongly inhibits Gram-positive bacteria and limits the swarming of Proteus, allowing for better colony definition. The differentiation is dual-fold: Bromothymol Blue acts as a pH indicator (turning yellow when lactose is fermented), and the addition of TTC (Triphenyltetrazolium Chloride) allows for the identification of organisms that reduce TTC to red formazan. E. coli and coliforms typically appear as yellow colonies with a yellow halo, while non-lactose fermenters remain blue or green.
