TSYEA Agar (Tryptone Soy Yeast Extract Agar) ISO is a nutrient-rich medium used for the confirmation of Listeria monocytogenes and other Listeria species. Formulated in strict accordance with the ISO 11290 standard, it is the primary medium used for the subculturing of presumptive colonies from selective isolation agars (such as Oxford or Palcam) before further biochemical and serological testing.

This medium is a modification of Tryptic Soy Agar (TSA) supplemented with Yeast Extract to provide a superior nutrient environment. The Pancreatic Digest of Casein and Soy Peptone provide organic nitrogen, amino acids, and peptides. Yeast Extract is added to provide a high concentration of B-complex vitamins, which are essential for the robust growth of Listeria. Sodium Chloride maintains the osmotic equilibrium, while Dipotassium Phosphate acts as a buffering agent to maintain the pH. TSYEA is non-selective, designed specifically to allow the pathogen to express its natural morphology and purity, ensuring that subsequent tests (such as Gram staining, motility, and the CAMP test) are performed on healthy, characteristic cultures.
