TSA Letheen Agar (Microbial Content Test Agar) is a highly nutritious medium used for the detection and enumeration of microorganisms in cosmetic products. It is specifically formulated to neutralize the antimicrobial activity of quaternary ammonium compounds and other preservatives, ensuring the recovery of bacteria from treated samples.

This medium is based on the formula of Tryptic Soy Agar (TSA) but is supplemented with neutralizing agents. The Pancreatic Digest of Casein and Soy Peptone provide a rich source of nitrogen, vitamins, and amino acids to support the growth of a wide variety of fastidious and non-fastidious microorganisms. The key feature of this medium is the addition of Lecithin and Polysorbate 80 (Tween 80). Lecithin acts as a neutralizer for quaternary ammonium compounds, while Polysorbate 80 neutralizes phenols, hexachlorophene, formalin, and, in combination with lecithin, ethanol. This makes the medium essential for "Microbial Content Tests" where the presence of disinfectant or preservative residues might otherwise cause false-negative results by inhibiting viable cells.
