Alkaline Saline Peptone Water ISO is a selective enrichment medium used for the detection and isolation of Vibrio species, including Vibrio cholerae and Vibrio parahaemolyticus, from food, water, and environmental samples. This medium is formulated in accordance with the ISO 21872 standard for the detection of potentially enteropathogenic Vibrio species.

This medium leverages the unique ability of Vibrio species to tolerate and thrive in environments with high alkalinity and elevated salt concentrations, which typically inhibit most other competing bacteria. The Enzymatic Digest of Casein (Peptone) provides the necessary nitrogen, vitamins, and amino acids for bacterial growth. The high concentration of Sodium Chloride (20.0 g/L) provides the halophilic conditions required by many Vibrio species. The alkaline pH (8.6 ± 0.2) acts as a selective agent, as most other commensal microorganisms found in food or water cannot grow at such high pH levels. This enrichment step is crucial for increasing the population of Vibrio prior to subculturing onto selective solid media like TCBS Agar.
