Eugon LT 100 Agar is a nutrient-rich medium specifically designed for the detection and enumeration of aerobic mesophilic bacteria in cosmetic products. It is formulated to overcome the inhibitory effects of preservatives commonly found in cosmetics, ensuring an accurate microbial count.
This medium is an adaptation of Eugon Agar, enhanced with neutralizing agents to facilitate the recovery of microorganisms from samples containing antimicrobial preservatives. It contains Pancreatic Digest of Casein and Papainic Digest of Soy Bean, which provide essential nitrogen, vitamins, and amino acids. Glucose serves as the primary energy source, while L-Cystine and Sodium Sulfite are included to stimulate the growth of more fastidious bacteria. The "LT" in the name refers to Lecithin and Polysorbate 80 (Tween 80); these two components work together to neutralize various inhibitory substances like quaternary ammonium compounds, phenolics, and aldehydes, allowing any viable bacteria present in the cosmetic sample to grow without interference.
Advantages
- Cosmetic Industry Standard: Formulated in accordance with ISO 21149 for the enumeration and detection of aerobic mesophilic bacteria.
- Effective Neutralization: Built-in lecithin and polysorbate 80 effectively counteract the preservative systems used in modern cosmetic formulations.
- Luxuriant Growth: Supports high-density (eugonic) growth of a wide range of bacteria, including Staphylococcus aureus and Pseudomonas aeruginosa.
- Versatile Detection: Suitable for use with various inoculation techniques, including pour-plate, spread-plate, and membrane filtration.
- High Sensitivity: Capable of recovering sublethally injured microorganisms that might otherwise be inhibited by residual sample toxicity.
Instructions for use
Suspend 46.4 g of the powder in 1 liter of distilled or deionized water. Mix thoroughly and heat with frequent agitation; boil for one minute to ensure complete dissolution. Preparation: Sterilize by autoclaving at 121 °C for 15 minutes. Note: Cool the medium to 45–50 °C before pouring. If using the membrane filtration method, immediately transfer the filtered membrane to the agar surface. Inoculate and incubate at 30–35 °C for 24 to 72 hours. Count the colonies and calculate the CFU per gram or mL of the original sample.
