Rothe Broth, also known as Glucose Broth with Azide, is a selective liquid medium used for the detection and enumeration of Enterococci in water, wastewater, food, and other environmental samples. It is primarily utilized in the presumptive phase of the Multiple Tube Fermentation technique to assess fecal contamination.
Rothe Broth is specifically formulated to allow the growth of Enterococci while effectively inhibiting competitive Gram-negative bacteria. Tryptone and Beef Extract provide the essential nitrogen, vitamins, and minerals required for bacterial proliferation. Glucose (Dextrose) serves as the fermentable carbohydrate and primary energy source. The selective action of the medium is due to the inclusion of Sodium Azide, which inhibits the cytochrome oxidase enzyme system of Gram-negative bacteria, thereby suppressing their growth. Sodium Chloride maintains the osmotic equilibrium. This medium is highly effective for the recovery of small numbers of Enterococci from samples with high levels of background microbial flora.
