Rogosa SL Agar is designed with a low pH and high salt concentration to create an environment that is selective for lactobacilli. Tryptic Digest of Casein and Yeast Extract provide the essential nitrogen, amino acids, and B-complex vitamins required for growth. The medium is enriched with three fermentable carbohydrates—Glucose, Arabinose, and Saccharose—which serve as diverse energy sources for the different metabolic pathways of various Lactobacillus species. Selectivity is primarily achieved through the high concentration of Glacial Acetic Acid and Sodium Acetate, which combined with a low pH (5.4), effectively inhibit the growth of streptococci, staphylococci, and most other commensal organisms. Manganese Sulfate, Magnesium Sulfate, and Ferrous Sulfate provide essential ions and growth factors that stimulate the proliferation of lactobacilli while maintaining the osmotic balance of the medium.
