Yeast Mold Broth (YM Broth) is a liquid medium used for the cultivation and enrichment of yeasts, molds, and other acid-tolerant microorganisms. Based on the formulation described by Wickerham, it is widely utilized in the food and beverage industries, as well as in molecular biology for the propagation of yeast cultures and the screening of fungal contaminants.
YM Broth provides a nutrient-rich liquid environment designed to maximize the biomass of fungal cultures. The Peptone and Yeast Extract serve as primary sources of nitrogen, amino acids, and essential B-complex vitamins. Malt Extract contributes additional carbohydrates (primarily maltose) and growth factors that are particularly effective for the rapid proliferation of fungi. Dextrose (Glucose) is included at a high concentration to provide an immediate energy source. The combination of these ingredients supports the growth of even the most fastidious yeasts and molds. Like the agar version, this broth can be rendered selective against bacteria by lowering the pH through the addition of organic acids after sterilization.
