The medium provides a balanced supply of nitrogen, vitamins, and energy to support robust fungal growth. Peptone serves as a source of nitrogen and amino acids, while Yeast Extract provides essential B-complex vitamins and growth factors. Malt Extract is particularly beneficial for fungi as it contains high concentrations of maltose and other saccharides. Dextrose (Glucose) acts as the primary fermentable carbohydrate and energy source. The relatively high concentration of dextrose, combined with the medium's naturally slightly acidic pH, favors the development of fungi over many bacterial species. Bacteriological Agar is the solidifying agent. The medium can be made highly selective by further acidification (lowering the pH to 3.0–4.0) or by the addition of antibiotics.
Advantages
- Broad-Spectrum Recovery: Effectively supports the growth of most yeasts and molds, including fastidious environmental strains.
- Wickerham Formulation: Follows the globally recognized standard for the maintenance and isolation of yeast cultures.
- Customizable Selectivity: The pH can be adjusted after sterilization to inhibit bacterial background flora, making it ideal for testing contaminated samples.
- Versatile Industrial Use: Widely used in the brewery and beverage industries for the detection of "wild" yeasts and spoilage molds.
- Clear Morphology: Promotes characteristic colony development, aiding in the preliminary identification and enumeration of fungal species.
