ESTY Broth is a liquid medium used for the cultivation and enrichment of diverse microorganisms, specifically optimized for the recovery of bacteria in food and environmental samples. It is formulated to provide a highly nutritious environment that supports the growth of fastidious species while maintaining metabolic stability.
ESTY Broth (also known as Esculin Sodium Thallite Yeast Broth in some specialized applications) is designed to act as a robust enrichment base. It contains Tryptone and Yeast Extract, which provide an abundance of nitrogenous compounds, B-complex vitamins, and essential growth factors. Dextrose is included as the primary fermentable carbohydrate to provide a rapid energy source for microbial proliferation. The medium is balanced with Sodium Chloride to maintain osmotic pressure. A key feature of this formulation is its buffering capacity, provided by Disodium Phosphate, which helps prevent excessive acidification during the fermentation of dextrose, thereby extending the viability of the culture during longer incubation periods.
Advantages
- High Nutrient Density: Supports the growth of a wide range of bacteria, including those with demanding nutritional requirements.
- Effective Enrichment: Ideal for reviving stressed or sublethally injured cells from processed food products.
- Metabolic Stability: The phosphate buffering system protects the population from rapid pH drops, ensuring higher final cell densities.
- Versatile Application: Can be used as a general enrichment broth or as a base for specific diagnostic tests when supplemented with selective agents.
- Reliable Performance: Consistent peptone quality ensures reproducible growth rates and metabolic patterns.
Instructions for use
Suspend 27.5 g of the powder in 1 liter of distilled or deionized water. Mix thoroughly until the powder is completely dissolved. Preparation: Distribute into appropriate containers (test tubes or flasks). Sterilize by autoclaving at 121 °C for 15 minutes. Note: For the isolation of specific pathogens, selective supplements may be added to the cooled sterile broth. Inoculate the sample into the broth and incubate at 35 ± 2 °C for 18–48 hours, or according to the specific protocol for the target microorganism. Observe for turbidity as an indicator of growth.
