LCS349 Cuvette test measures 0.01-0.5 mg/L PO₄-P, 0.03–1.50 mg/L PO₄ and 0.02–1.20 mg/L P₂O₅ in wastewater, drinking water and other liquids. The product uses phosphormolybdenum blue (ISO 6878-1-1986, DIN 38405 D11-4). Article LCS349 can be used for 25 tests. The product is compatible with Hach spectrophotometers DR3900 and DR6000. LCS349 reagent kit is intended to be used together with 50mm semi-micro cuvettes (LZP341).

Orthophosphate is measured without hydrolysis. Total phosphorus requires hydrolysis in order to be analysed. See LCS349 Cuvette test manual for more information regarding each procedure. During the reaction phosphate ions are reacting with molybdate and antimony ions in an acidic solution in order to form an antimonyl phosphormolybdate complex that is reduced by ascorbic acid to phosphormolybdenum blue. The water sample pH is required to be between 2-10 pH. The temperature of the water sample and reagents should be 15-25 °C (59-77 °F). Lower and higher temperatures may cause low-bias results.

For short-term sample conservation of the wastewater, drinking water or other liquid sample the preparation is not required. For longer-term preservation it is recommended to reduce the pH of the sample with sulphuric acid to pH<2. The water sample should be kept in glass, polyethylene or borosilicate vessel. Abovementioned recommendations refer only to the total phosphorus analysis. The storage conditions of the LCS349 reagent set are between 15 °C and 25 °C.
