Originally developed by De Man, Rogosa, and Sharpe in 1960, MRS Broth is an enriched medium that provides all the essential nutrients for fastidious lactic acid bacteria. The formulation includes peptones and yeast extract as sources of nitrogen, vitamins, and minerals. Dextrose (glucose) serves as the primary fermentable carbohydrate. The addition of Tween® 80 (polysorbate 80), magnesium sulfate, and manganese sulfate provides essential growth factors and ions. Sodium acetate and ammonium citrate act as selective agents, inhibiting the growth of most accompanying flora, including gram-negative bacteria and many fungi, while supporting the growth of Lactobacilli even at lower pH levels.
