This "Modified" version of the classic Terrific Broth provides a sophisticated nutrient balance to prevent the common pitfalls of high-density bacterial growth. It contains a high concentration of Tryptone and Yeast Extract, providing an abundance of amino acids, vitamins, and trace elements. Unlike standard LB broth, Modified Terrific Broth is heavily buffered with Potassium Phosphates to prevent the acidification of the medium—a result of metabolic byproduct accumulation—which often leads to premature cell death. The formulation is specifically designed to be used with Glycerol as the carbon source instead of Glucose; this prevents catabolite repression and avoids the rapid buildup of acetic acid, allowing the culture to reach a significantly higher biomass.
