Triple Sugar Iron Agar (TSI) is a differential medium used for the identification of Enterobacteriaceae based on the fermentation of specific carbohydrates and the production of hydrogen sulfide. It is a fundamental tool in diagnostic microbiology for differentiating Gram-negative enteric bacilli, particularly Salmonella and Shigella, from other coliforms.

The medium is designed to test for the fermentation of three sugars: Glucose, Lactose, and Sucrose. It contains Beef Extract, Yeast Extract, and Peptone Mixture as sources of nitrogen, vitamins, and minerals. Phenol Red serves as the pH indicator, which turns yellow in the presence of acid (fermentation) and remains red/pink under alkaline conditions. Ferrous Ammonium Sulfate and Sodium Thiosulfate are included to detect the production of Hydrogen Sulfide (H₂S); if H₂S is produced, it reacts with the iron salt to form a black precipitate. The medium is prepared as a "slant," allowing for both aerobic (slant) and anaerobic (butt) growth, providing a comprehensive metabolic profile of the organism in a single tube.
